Abstract |
Background s: The mesangial extracellular ma- trix expansion appear to be important features of progressive glomerular diseases and the mononuclear cells may play a pivotal role in this process. Method: We evaluated the effects of superna- tants of peripheral blood mononuclear cells (PBMCS) activated by concanavalin A on the expression of VCAM-1 and ICAM-1, the secretion of monocyte chemotactic protein-l(MCP-l), the thymidine incor poration, and the fibronect'@in production in MC by {3H}-thymidine incorporation, flow cytometry, or enzyrne immunoassay. Results 1) Expression of VCAM-1 and ICAM-1 by MC was enhanced by 43% and 53%, respectively, after the addition of PBMCS after 24 hours, as compared with media(P<0.001). 2) Secretion of MCP-1 was not different between addtion of PBMCS and media alone. 3) PBMCS inhibited MC thymidine incorporation by 59%, as compared with media alone(P<0.001). 4) PBMCS enhanced the fibronectin secretion in MC by 36% after 96 hours, as compared with media alone(P<0.05). Conclusion: PBMCS enhanced expression of VCAM-1 and ICAM-1 by MC which stimulated leukocyte infiltration into glomeruli. And PBMCS inhibited the thymidine incorporation and enhanced the fibronectin secretion by MC, leading to mesangial expansion and eventually glomerulo- sclerosis. |
