고양이 신장에서 Ethacrynic Acid의 이뇨효과에 대한 Probenecid 의 영향

Abstract
Effect of probenecid on the diuretic response of etha- crynic acid was studied by clearance method in the cat. Ethacrynic acid was infused through the femoral vein before or after the treatment of probenecid. 1) Ethacrynic acid increased the urine flow and fractioal Na excretion (FE,) with doses 0.5 to 25 mg/kg as the dose was progressively raised, but decreased glomeru lar filtration rate (GFR) with the dose range of 1 to 25 mg/kg. Increase in fractional K excretion (FE ,) was maximal at a dose of 5 mg/kg. 2) During the continous infusion of ethacrynic acid (5 mg/kg-6 mg/kg/h), the administration of probenecid (30 mg/kg-36mg/kg/h) failed to block the natriuretic and kaliuretic effects of ehacrynic acid. 3) The maximal response of ethacrynic acid (5 mg/ kg) was not changed by the pretreatment of probenecid (30 mg/kg-36 mg/kg/h), indicating that the pretreat- ment of probenecid did not influence the diuretic effect of ethacrynic acid. 4) The pretreatment of probenecid (30mg/kg-36 mg/kg/h) failed to abolish the diuretic effect of fur- osemide (3 mg/kg). 5) Ethacrynic acid (5 mg/kg-6 mg/kg/h) reduced significantly CpAg and TpAg. These results suggest that ethacrynic acid inhibited both clearance and tubular secretion of PAH, but its diuretic response was not affected by probenecid in cat kidney.
